You could put a password on the epub file using a zip program, I suppose, but it wouldn't be terribly useful for distribution.
I think you might be in the wrong forum, though. Are you asking about DRM and encryption? Is there a corollary accessibility question you were wondering about?
It's better not to apply DRM to content if you can, but if you do, don't prevent access to the text from screen readers, refreshable braille devices, etc.

If you put a password on the ZIP file, it will not be a valid EPUB file since the mimetype must be readable outside the ZIP file.
The only way is to use DRM and as said before it is better not to use it.
The EPUB format does not have built in support for DRM.
