I am looking for advise on possible solution and what tags need to be changed for Manga ePub file to enable right to left page turns on specifically eReader devices like iPad and Nook Color.

The bad news is I'm not sure that this is doable now on iPad or Nook Color. To glean information about feature support details in today's eReaders, I suggest a twitter post with #ePrdctn (maybe also @lizcastro).
The good news is that the latest version of EPUB, EPUB 3, which is being adopted during this year, has full support for global languages including page-progression-direction. See: http://idpf.org/epub/30/spec/epub30-publications.html#sec-spine-elem .
